Collective Cell Behaviors During Skin Regeneration

Abstract:

Hair follicle is a prototypical model system for studying principles and mechanisms of stem cell control in the context of tissue growth and regeneration. Notably, each hair follicle grows in a highly stereotypical manner—the hair growth cycle, and on the entire skin, cyclical growth of many thousands of hair follicles is coordinated via a self-organized process, whose outcome are the so-called hair growth waves (Refs 1, 2). Periodic activation of epithelial stem cells is the critical pre-requisite of hair growth, and for this reason their activation status is tightly regulated by a number of paracrine signals emanating both from their immediate niche cells inside of the follicle (Ref 3), as well as from distantly positioned non-hair skin cells (Ref 4). Hair stem cells activate when the collective signaling input that they receive from all sources definitively reaches stable above- the-threshold level. In the lecture, current state of knowledge on the cellular and molecular components of the long-range niche signals that drive collective hair growth and their intersection with the bona fide hair follicle niche counterparts (Ref 3, 4) as well as the application of single cell bioinformatics to studying these tissue-level questions (Ref 5) will be discussed. Knowledge gleaned from studying collective growth by hair follicles in the skin can reveal general principles that likely operate in other tissues to achieve coordinated regeneration of their respective stem cell-rich units (Ref 6).
